General
SensEye® is the evolving sensor program for label-free, multiplex analysis. SensEye® sensors are designed exclusively for use in the MX96 imaging Surface Plasmon Resonance instrument from IBIS Technologies.
Each SensEye type expresses a specific functionality for immobilization or capturing of ligand molecules such as antibodies. SensEyes are available with two different architectures of their sensing surface: sensors with a planar surface (P-type) and sensors with a 2-dimensional surface (G-type). P-type SensEyes are preferably used for applications in which kinetics are more important than sensitivity, whereas G-type SensEyes are recommended for applications where sensitivity is the larger challenge. For both SensEye® types, the chemical/biological functionality is present in a bio-inert background, thus ensuring a minimal non-specific interaction.
SensEyes are available with different surface chemistries, divided into three main categories: surfaces for covalent ligand immobilization (Basic SensEyes), surfaces for direct ligand capturing (Antibody Capturing SensEyes) and surfaces for capturing of tagged ligands (Tagged Ligand Capturing SensEyes). Thanks to the bio-inert background surface of the SensEyes, ligands can also be captured from crude samples, which contributes to a comfortable use of these sensors.
Basic SensEyes
Basic SensEyes are applied for covalent binding of ligands, for instance, via carbodiimide chemistry. Chemically active SensEyes such as the Easy2Spot can be used for direct, covalent ligand immobilization without the need of preceding surface chemistry. In combination with a spotting device like the Continuous Flow Micro-spotter, an array of many different ligands can be spotted on these SensEyes.
Features:
- Stable covalent immobilization.
- Easy to use pre-activated surfaces.
- Can be combined with array spotting for cost effective multiplex analysis.
Antibody Capturing SensEyes
The antibody capturing SensEyes capture antibodies through their Fc-moiety, resulting in sensor surfaces on which the Fab region of the antibodies is optimally exposed for interaction analysis. In combination with a spotting device such as the Continuous Flow Micro-spotter an array of many different antibodies can be spotted on these SensEyes.
Features:
- Oriented antibody immobilization.
- Compatible with FixIt for irreversible immobilization of antibodies after capturing.
- Antibody capturing from crude samples.
Tagged Ligand Capturing SensEyes
Tagged ligand capturing SensEyes bind ligands via the tag that is present on these ligands. Examples of suitable tags are biotin and histag. The high affinity of these SensEyes allow ligand capturing from crude samples with low ligand concentrations. In combination with a spotting device such as the Continuous Flow Micro-spotter an array of many different antibodies can be spotted on these SensEyes.
Features:
- High affinity surfaces allowing ligand capturing from diluted samples.
- Ligand capturing from crude samples.
- Low a-specific interaction.
